General description
X-Gal, or 5-bromo-4-chloro-3-indolyl-β-D-galactopyranoside, is an organic compound commonly used in molecular biology as a chromogenic substrate for the enzyme β-galactosidase, which is encoded by the LacZ gene. When β-galactosidase hydrolyzes X-Gal, it forms galactose and 5,5′-dibromo-4,4′-dichloro-indigo, resulting in the formation of intense blue color colonies. This process is widely used in LacZ activity assays and staining techniques, such as X-gal staining or LacZ staining to distinguish between recombinants (white) and nonrecombinants (blue). X-Gal staining is frequently used in the fields of molecular biology, genetics, and developmental biology to study gene expression and cell line tracing. X-Gal Solution is provided as a convenient 40 mg/ml concentrate in DMSO, ready for dilution into the culture medium or appropriate buffers.
Application
X-Gal Solution - Novagen has been used at 1:40 dilution to detect the expression of β-galactosidase and mutant fusion protein generated by the ApcΔ242 mutation in the tissue sample.
