Specificity

Clone IL11 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Interferon regulatory factor 5 (IRF-5). It targets an epitope within 19 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 19 amino acids from the N-terminal half of human Interferon regulatory factor 5 (IRF-5).

Application

Quality Control Testing

Evaluated by Western Blotting in LNCap cell nuclear extract.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected IRF-5 in LNCap cell nuclear extract.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected IRF-5 in THP-1 and NIH3T3 cell nuclear extracts.

Flow Cytometry Analysis: 0.1 μg from a representative lot detected IRF-5 in one million THP-1 cells.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected IRF-5 in THP-1 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected IRF-5 in human tonsil tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound IRF-5 peptide with a KD of 4.9 x 10-7 in an affinity binding assay.

Target description

Interferon regulatory factor 5 (UniProt: Q13568; also known as IRF-5) is encoded by the IRF5 gene (Gene ID: 3663) in human. The interferon regulatory factors (IRFs) are a family of nine transcription factors that regulate pathogen-induced innate and acquired immune responses. In unstimulated cells they reside in the cytoplasm. IRF-5 plays a critical role in innate immunity by activating expression of type I interferons (IFN- and β) and inflammatory cytokines downstream of endolysosomal toll-like receptors TLR7, TLR8, and TLR9. It regulates the transcription of IFN genes (IFNA and IFNB) and IFN-stimulated genes (ISG) by binding to an interferon-stimulated response element (ISRE) in their promoters. It is shown to be a key transcription factor regulating the interferon response during SARS-CoV-2 infection. It is maintained as a monomer in an autoinhibited state and undergoes homodimerization and activation following its phosphorylation on serine 451 and 462 in the C-terminal autoinhibitory region. It has been reported that phosphorylation of serine 451 relieves the autoinhibitory conformation, and phosphorylation of serine 462 stabilizes the IRF-5 homodimer. In its phosphorylation and activation scheme, the innate adapter protein TASL (TLR adaptor interacting with SLC15A4 on the lysosome) recruits IRF-5, which is then phosphorylated by IKK-β at serine 462 that allows IRF-5 dissociation from the adapter protein followed by its dimerization and nuclear translocation. Mutations in IRF5 gene have been linked to systemic lupus erythematosus 10 (SLEB10), rheumatoid arthritis, and irritable bowel syndrome. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
