General description

pRc and pRSV Supercoiled Expression Vectors (5.2 kb) are for protein expression in mammalian cells. Each vector contains:
  • The polyadenylation signal and transcription termination sequence from BGH to enhance mRNA stability
  • The SV40 replication origin for episomal replication
  • The f1 origin for rescue of the sense strand
  • The neomycin resistance gene, and
  • An ampicillin resistance gene and colE1 origin for selection and maintenance in E. coli.


The primary difference between these expression vectors is that pRc contains the enhancer-promoter sequence from human cytomegalovirus (CMV2) and pRSV from Rous sarcoma virus (RSV). Both are strong promoters for high-level transcription.

The pRc MCS contains unique ApaI, HindIII, NotI, and XbaI restriction sites, plus redundant BstXI sites.

The pRSV MCS contains unique HindIII, NotI, SpeI, and XbaI restriction sites, plus redundant BstXI sites.

Specificity

The MCS contains unique ApaI (CMV2 only), HindIII, NotI, SpeI (RSV only), and XbaI restriction sites, plus redundant BstXI sites.
