Cell Line Description

Established from a NIH Swiss mouse embryo. These cells are sensitive to sarcoma virus focus formation and leukemia virus propogation. Cells have now lost their contact inhibition.

Application

DNA transfection studies
NIH 3T3 Cell Line has been used to:
  • study calcium-mediated actin reset (CaAR) in response to physiological changes
  • study the effects of activation of the transient receptor potential vanilloid 3 channel (TRPV3) on adipocyte differentiation
  • determine the viral titer of the retroviral construct -ST40hTNFα

Culture Medium

DMEM + 2mM Glutamine + 10% Calf Serum (CS).

Subculture Routine

Split sub-confluent cultures (70-80%) 1:2 to 1:6 i.e. seeding at 2-5x10,000 cells/cm2 using 0.25% trypsin/EDTA; 5% CO2; 37°C. Do not allow culture to become fully confluent; the use of fetal calf serum is not recommended.
