General description

Intended for the histochemical demonstration of neutrophil granules in blood or bone marrow films.

Principle

Following fixation, blood or bone marrow films are immersed in a buffered Sudan Black B solution. After rinsing, slides are counterstained with Gill hematoxylin, and cells are examined microscopically for the presence of blue-black discrete granulation. Granulocytic precursors demonstrate groups of coarse blue-black deposits. Other hematopoietic cells are typically negative.

Emergency granulopoiesis occurs in response to severe microbial infection. However, whether and how other blood components, particularly monocytes/macrophages and their progenitors, including hematopoietic stem/progenitor cells (HSPCs), participate in the process and the underlying molecular mechanisms remain unknown.
