General description

Tau-441 human, recombinant, is an isoform of Tau, variant 2N4R, having 4 microtubule binding repeats (R) and 2 amino terminal inserts (N). Tau is a family of neuronal microtubule-associated proteins. Tau, a phosphoprotein is considered as an important microtubule associated protein (MAP) of a mature neuron. The 2N4R tau consists of 441 amino acids (tau441) and is considered as the largest size human brain tau. In human brain, tau is present in six molecular isoforms.
The gene microtubule associated protein tau (MAPT) is localized on human chromosome 17q21.3. It is expressed in neurons, more specifically in axons.

Application

Human Tau-441 has been used to study the effects of tau on neuropathological changes in Alzheimer′s disease.
Tau-441 human has been used to study its effects on mitochondrial transport velocities found in glial cells.

Biochem/physiol Actions

Microtubule associated protein tau (MAPT) plays an important role in the assembly and maintenance of the microtubule structure. Loss of expression of MAPT results in developmental delay and learning disability. The protein has been associated with pathology of Alzheimer′s disease (AD).
Tau promotes assembly and stabilizes neuronal microtubules under normal physiological conditions.

Reconstitution

Lyophilized from MES, pH 6.8, containing NaCl and EGTA. When reconstituted in water to a protein concentration of 1 mg/mL, the resulting buffer will have ~50 mM MES, pH 6.8, 100 mM NaCl, and 0.5 mM EGTA.
