General description

Currently, there are 13 caspases recognized in mammals. Caspases are classified as initiators and effectors. Caspase 8 belongs to the class of initiators. It possesses a prodomain at the terminal and contains the death effector domain which serves as protein-protein interaction motif for caspase 8. With respect to substrate specificity, caspase 8 is classified as a member of group III. It specifically recognizes the (I/L/V) EXD sequence on the substrate.

Application

Caspase Inhibitor has been used to study the specificity of the CIL-1 (desglucoanagalloside B) interaction with the caspase 3/7 system.

Biochem/physiol Actions

The peptide Ac-Ala-Glu-Val-Asp-al, or Ac-AEVD-CHO, is an inhibitor of Group III caspases. It acts upon caspases-6, 8, 9, and 10, with particular effectiveness on caspase-6 and caspase-8. The use of Ac-AEVD-CHO in several apotosis studies has been reported.
