General description

Mammals contain variants of histone H3, that are, the replicative histones (H3.1 and H3.2) and the replacement histones (H3.3). Histone H3 is located on human chromosome 6p22.

Immunogen

The epitope recognized by PLA0148 maps to a region between residue 100 and the C-terminus (residue 135) of human Histone H3 using the numbering given in entry NP_003520.1 (GeneID 8350).

Biochem/physiol Actions

Histone proteins are the basic building blocks of chromatin. Histone H3 controls protein-protein interactions to induce binding of trans-acting factors that have a role in chromatin condensation. Histone H3 K27M mutations are observed in adult cerebellar high-grade gliomas.

Other Notes

Histone H3 is one of four histone proteins that assemble into an octamer to form the nucleosome core and serve to spool and compact DNA into chromatin. Histones are subject to a complex set of post-translational modifications (acetylation, methylation, phosphorylation, ubiquitination, sumoylation, citrullination, and ADP ribosylation) that are important to the regulation of gene transcription, DNA replication, and DNA repair.
