General description

Protein FAM83B (UniProt: Q5T0W9; also known as Family with Sequence Similarity 83, member B) is encoded by the FAM83B (also known as C6orf143) gene (Gene ID: 222584) in human. FAM83B is a member of an 8-member protein family (FAM83) that shares the highly conserved DUF1669. It is a proto-oncogene that functions in the epidermal growth factor (EGF) receptor signaling pathway. May activate both the EGFR itself and downstream RAS/MAPK and PI3K/AKT/TOR signaling cascades. It interacts with RAF1 leading to displacement of 14-3-3 proteins from RAF1, which then activates RAF1 within the RAS/MAPK signaling cascade. Higher levels of FAM83B are shown to increase PI3K/AKT signaling and cell proliferation. In EGFR-dependent breast cancer cells that express higher levels of FAM83B expression, ablation of FAM83B is shown to diminish p110 alpha and AKT membrane localization, which result in reduced AKT phosphorylation and cell proliferation.
