General description
Bicelle kit contains 200mg of dimyristoylphosphatidylcholine (DMPC), 200mg of dimyristoylphosphatidylglycerol (DMPG), 1g of 3-[(3-cholamidopropyl)-dimethylammonio]-1-propane sulfonate (CHAPS) and 1g of 3-(cholamidopropyl)dimethylammonio-2-hydroxy-1-propane-sulfonate (CHAPSO). Bicelle is a discoidal lipid aggregate. A bicelle consists of a planar bilayer at the center made of long-chain phospholipids, which are encircled by a detergent layer. The planar bilayer is generally composed of DMPC, which may have phospholipids such as (DMPG) for alteration of charge of the membrane. The detergent layer is composed of bile salt derivative, such as CHAPSO.
The bicelle kit is designed to aid with the production of high-quality membrane protein crystals. Bicelles can be integrated into standard crystallization protocols, and in contrast to micelles, bicelles maintain the protein in a more native bilayer environment allowing proteins to be captured in a more biologically relevant orientation.
